What is Pharmacometrics?


Pharmacometrics is a field of study that uses modeling and simulation to quantify the relationship between the administered drug and its interaction with the body's response. Mathematical and statistical methods are utilized to develop models which are often constructed based on the underlying pharmacology, physiology or disease.  Pharmacometric analyses have become common in the pharmaceutical industry as they are important for decision making in drug development, regulatory expectations and drug product labeling. The Importance of PK/PD


Pharmacokinetic/Pharmacodynamic (PK/PD)  are important components of pharmacology. It is commonly said that PK describes what the drug does to the body while PD describes what the body does to the drug. In other words, PK describes what happened to the drug molecule once enters and resides in the body, i.e. how it is absorbed into the body (absorption), how it breaks down (metabolism), how it spread throughout the body (distribution), and how it leaves the body (excretion).  PD, on the other hand, describes how the body respond to the drug. For example, lower heart rate is a PD response after taken propranolol.  PK and PD can be integrated by using mathematical relationships. Once the relationship is established, drug developers can use it to select the dose for future clinical studies. In addition, results from PK/PD analyses can be used to help design clinical studies and contribute to drug product labeling.
